Samples were incubated in CD36 polyclonal antibody (Product # PA1-16813) using a dilution of 2 µg/mL followed by an anti-rabbit HRP secondary antibody. Separated on a 12% gel by SDS-PAGE, transferred to PVDF membrane and blocked in 5% non-fat milk in TBST. Detection: chemiluminescence.

- Western Blot was performed using Anti-CD36 Polyclonal Antibody (Product # PA1-16813) and a 88 kDa band corresponding to glycosylated form of Platelet glycoprotein 4 was observed in 3T3-L1 upon differentiation to adipocytes. Also, the protein expression was observed to be higher in adipose and brown adipose tissues when compared to other tissues tested. Membrane enriched extracts (30 µg lysate) of 3T3-L1 (Lane 1), 3T3-L1 differentiated to adipocytes (Lane 2), Mouse Adipose (Lane 3), Mouse Brown Adipose (Lane 4), Rat Adipose (Lane 5), Rat Brown Adipose (Lane 6), Mouse Brain (Lane 7), Rat Brain (Lane 8), Mouse Kidney (Lane 9), Mouse Liver (Lane 10) and Rat Testis (Lane 11) were electrophoresed using NuPAGE™ 4-12% Bis-Tris Protein Gel (Product # NP0322BOX). Resolved proteins were then transferred onto a Nitrocellulose membrane (Product # IB23002) by iBlot® 2 Dry Blotting System (Product # IB21001). The Blot was probed with the primary antibody (1:1000 dilution) and detected by chemiluminescence with Goat anti-Rabbit IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036, 1:6000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005).

- Figure 6 CD36 expression in normal (HEKa) and dysplastic (DOK) keratinocytes after UVA irradiation. Expression of CD36 in both HEKa and DOK cells was assessed by confocal immunofluorescence and confirmed by Western blot. In basal conditions, CD36 expression in normal keratinocytes is undetectable with the selected methods. Its expression increases in dysplastic cells, as well as following UVA irradiation.
