- Western blot analysis of MRE11 in 0.5 mg/mL HeLa lysate. Samples were incubated in MRE11 polyclonal antibody (Product # PA3-16527). A band appears at the theoretical molecular weight of 81 kDa for this product. This experiment was performed under reducing conditions using the 12-230 kDa separation system.

- Knockdown of MRE11 was achieved by transfecting HeLa cells with MRE11 specific siRNAs (Silencer® select Product # s8959, s8960). Western blot analysis (Fig. a) was performed using whole cell extracts from the MRE11 knockdown cells (lane 3), non-specific scrambled siRNA transfected cells (lane 2) and untransfected cells (lane 1). The blots were probed with MRE11 Polyclonal Antibody (Product # PA3-16527, 1:5000 dilution) and Goat anti-Rabbit IgG (H+L) Superclonal™ Secondary Antibody, HRP conjugate (Product # A27036, 0.25 µg/ml, 1:4000 dilution). Densitometric analysis of this western blot is shown in histogram (Fig. b). Decrease in signal upon siRNA mediated knock down confirms that antibody is specific to MRE11.

- Western blot analysis was performed on whole cell extracts (30 µg lysate) of HeLa (Lane 1), SKNAS (Lane 2), K562 (Lane 3), Jurkat (Lane 4), Raji (Lane 5), A431 (Lane 6), HepG2 (Lane 7) and IMR-32 (Lane 8). The blot was probed with Anti-MRE11 Polyclonal Antibody (Product # PA3-16527, 1:3000 dilution) and detected by chemiluminescence using Goat anti-Rabbit IgG (H+L) Superclonal™ Secondary Antibody, HRP conjugate (Product # A27036, 0.25 µg/ml, 1:4000 dilution). An 81kDa band corresponding to MRE11 was observed across all cell lines tested.
