- PA1-125 recognizes MnSOD in bovine, human, mouse, and rat samples and has been used in WB, and IHC(P) applications. This antibody was originally validated as part of a Thermo Scientific Cellomics High Content Screening Kit. The antibody sold separately may have slightly different performance and may need to be further optimized for the best results.

- Western blot analysis of MnSOD was performed by loading 25 µg of SH-SY5Y (lane 1), HepG2 (lane 2) and mouse liver (lane 3) onto an SDS polyacrylamide gel. Proteins were transferred to a PVDF membrane and blocked at 4ºC overnight. The membrane was probed with a MnSOD polyclonal antibody (Product # PA1-125) at a dilution of 1:1000 overnight at 4°C, washed in TBST, and probed with an HRP-conjugated secondary antibody for 1 hr at room temperature in the dark. Chemiluminescent detection was performed using Pierce ECL Plus Western Blotting Substrate (Product # 32132). Results show a band at ~21 kDa.

- Western blot analysis was performed on whole cell extracts (30 µg lysate) of A549 (Lane 1), A549 treated with TNF alpha (20ng/mL for 72 hrs) (Lane 2), THP-1 (Lane 3), HEK-293 (Lane 4), MCF7 (Lane 5) and T-47D (Lane 6). The blot is probed with SOD2 Polyclonal Antibody (Product # PA1-125, 1:500 dilution) and detected by chemiluminescence using Goat anti-Rabbit IgG (H+L) Superclonal™ Secondary Antibody, HRP conjugate (Product # A27036, 0.25 µg/ml, 1:4000 dilution). A band at 21kDa corresponding to SOD2 was observed across cell lines tested and enhanced upon TNF alpha treatment in A549 cell line.
