- PA1-043 detects Post Synaptic Density 93 (PSD-93) from rat and human tissues. This antibody does not detect other synapse-associated protein family members. PA1-043 has been successfully used in Western blot and immunohistochemistry procedures. By Western blot, this antibody detects an ~110 kDa protein representing PSD-93 in rat brain extract. Immunohistochemical staining of PSD-93 in rat cerebellum with PA1-043 yields a pattern consistent with post-synaptic localization. The PA1-043 immunogen is a synthetic peptide corresponding to residues N(352) K L C D K P A S P R H Y S P(366) of human PSD-93. This sequence is completely conserved in human PSD-93. PA1-043 immunizing peptide (Cat. # PEP-021) is available for use in neutralization and control experiments.

- Western blot analysis was performed on membrane-enriched extracts (30 µg lysate) of SHY-SY5Y (Lane 1), and Neuro-2a (Lane 2). The blots were probed with Anti-PSD93 Rabbit Polyclonal Antibody (Product # PA1-043, 3 µg/mL) and detected by chemiluminescence using Goat anti-Rabbit IgG (H+L) Superclonal™ Secondary Antibody, HRP conjugate (Product # A27036, 0.25 µg/mL, 1:4000 dilution). A 115 kDa band corresponding to PSD93 was observed across the cell lines tested. Known quantity of protein samples were electrophoresed using Novex® NuPAGE® 4-12 % Bis-Tris gel (Product # NP0321BOX), XCell SureLock™ Electrophoresis System (Product # EI0002) and Novex® Sharp Pre-Stained Protein Standard (Product # LC5800). Resolved proteins were then transferred onto a nitrocellulose membrane with iBlot® 2 Dry Blotting System (Product # IB21001). The membrane was probed with the relevant primary and secondary Antibody following blocking with 5 % skimmed milk. Chemiluminescent detection was performed using Pierce™ ECL Western Blotting Substrate (Product # 32106).
