Human plasma contains at least two
distinct forms of kininogen, the ca
rrier molecule of bradykinin, which
can be distinguished by their phys
ical, chemical and immunochemical p
roperties. They are designated high
-molecular-weight kininogen (HMWK)
and low-molecular weight kininogen
(LMWK). HMWK (MW 110,000) and LMWK
(MW 50,000) both liberate bradykini
n following the action of kallikrei
n. HMWK circulates as a non-enzymat
ic factor that is central to contac
t activation reactions. LMWK plays
no known role in the activation of
coagulation. Normal adult plasma le
vels are 7 mg/100 ml for HMWK as de
termined by electroimmunodiffusion
and 9 mg/100 ml based on haemagglut
ination inhibition assays and RIA.
Low plasma kininogen levels are see
n in newborn and pre-term infants.
HMWK is absent in congenital kinino
gen deficiency of the named affecte
d families Fitzgerald and Reid, Wil
liams and Flaujeac lack both HMWK a
nd LMWK. The condition is associate
d with the decreased fibrinolytic a
ctivity and kinin formation but is
asymptomatic. Purified kininogen fr
om pooled human plasma is used for
the immunization. Freund‰Ûªs com
plete adjuvant is used in the first
step of the immunization procedure
