Western blot was performed using Anti-DRAM1 Rabbit Polyclonal Antibody (Product # PA5-20335) and a 15 kDa band corresponding to DRAM1 was observed across cell lines tested except Raji and Jurkat. Membrane enriched extracts (30 µg lysate) of HCT 116 (Lane 1), HeLa (Lane 2), Hep G2 (Lane 3), MCF7 (Lane 4), Raji (Lane 5) and Jurkat (Lane 6) were electrophoresed using NuPAGE™ 12% Bis-Tris Protein Gel (Product # NP0342BOX). Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1 µg/mL) and detected by chemiluminescence with Goat anti-Rabbit IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036, 1:4000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005).

Immunohistochemical analysis of paraffin-embedded human liver tissue using DRAM Polyclonal Antibody (Product # PA5-20335) at 2.5 µg/mL. Tissue was fixed with formaldehyde and blocked with 0.1 serum for 1 h at RT; antigen retrieval was by heat mediation with a citrate buffer (pH6). Samples were incubated with primary antibody overnight at 4˚C. A goat anti-rabbit IgG H&L (HRP) at 1/250 was used as secondary. Counter stained with Hematoxylin.
