Western blot was performed using Anti-SOX9 Polyclonal Antibody (Product # PA1-29537) and a 56kDa band corresponding to SOX9 was observed in all the tested cell models, except THP-1 along with an uncharacterized band at ~50 kDa. Modified whole cell lysate (1% SDS) (30ug lysate) of THP-1 (Lane 1), Jurkat (Lane 2), Hep G2 (Lane 3) and PANC-1 (Lane 4) were electrophoresed using Novex® NuPAGE® 4-12 % Bis-Tris gel (Product # NP0322BOX). Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1µg/ml concentration) and detected by chemiluminescence with Goat anti-Rabbit IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036, 1:4000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using SuperSignal™ West Dura Extended Duration Substrate (Product # 34076).

Dot blot analysis of the SOX-9 peptide (pSer181) and SOX-9 peptide probed with the anti-SOX-9 (pSer181) (Product # PA1-30708) and anti-SOX-9 (Product # PA1-29537). The staining shows that the anti-SOX-9 (pSer181) recognized the SOX-9 peptide (pSer181) only; whole the anti-SOX-9 could bind both the SOX-9 peptide (pSer181) and SOX-9 peptide.
