- Knockdown of CRMP2 was achieved by transfecting Neuro-2a with CRMP2 specific siRNAs (Silencer® select Product # s4269). Western blot analysis (Fig. a) was performed using membrane enriched extracts from the CRMP2 knockdown cells (lane 3), non-specific scrambled siRNA transfected cells (lane 2) and untransfected cells (lane 1). The blot was probed with CRMP2 Monoclonal Antibody (OTI5D7) (Product # MA5-27498, 1:2000 dilution) and Goat anti-Mouse IgG (H+L), Superclonal™ Recombinant Secondary Antibody, HRP (Product # A28177, 0.25µg/ml, 1:4000 dilution). Densitometric analysis of this western blot is shown in histogram (Fig. b). Decrease in signal upon siRNA mediated knock down confirms that antibody is specific to CRMP2.

- Western blot was performed using Anti-CRMP2 Monoclonal Antibody (OTI5D7), eBioscience™ (Product # MA5-27498) and a 60 kDa band corresponding to CRMP2 was observed across cell lines and tissues tested except MCF-7 and SK-BR-3 which are reported to be low protein expressing cell lines. Membrane enriched extracts (30 µg lysate) of PC-12 (Lane 1), Neuro-2a (Lane 2) and SH-SY5Y (Lane 3), U-87 MG (Lane 4), MCF-7 (Lane 5), MCF 10A (Lane 6), SK-BR-3 (Lane 7), tissue extracts of Mouse Brain (Lane 8) and Rat Brain (Lane 9) were electrophoresed using NuPAGE™ 4-12% Bis-Tris Protein Gel (Product # NP0322BOX). Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1:2000 dilution) and detected by chemiluminescence with Goat anti-Mouse IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A28177, 1:4000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005).
