- Western blot was performed using Anti-Cathepsin H Polyclonal Antibody (Product # PA5-103115) and a 37 kDa band corresponding to pro-cathepsin H was observed in Hep G2 but not in Jurkat. Whole cell extracts (40 µg lysate) of Hep G2 (Lane 1), Hep G2 treated with 1X-PTI (Lane 2), Jurkat (Lane 3) and Jurkat treated with 1X-PTI (Lane 4) were electrophoresed using NuPAGE™ 4-12% Bis-Tris Protein Gel (Product # NP0322BOX). Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1:1000 dilution) and detected by chemiluminescence with Goat anti-Rabbit IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036,1:20,000) using the iBright™ FL1500 Imaging System (Product # A44115). Chemiluminescent detection was performed using SuperSignal™ West Dura Extended Duration Substrate (Product # 34076). Hep G2 is a high-expressing cell line for CTSH but Jurkat is a low-expressing cell line. Upregulation was observed in Hep G2 upon treatment with protein transport inhibitor. An uncharacterized band (*) was observed around ~90 kDa.

- Immunofluorescent analysis of Cathepsin H in HeLa cells. Samples were fixed with paraformaldehyde, permeabilized with 0.1% Triton X-100, blocked with 10% serum (45 min at 25°C) incubated with Cathepsin H polyclonal antibody (Product # PA5-103115) using a dilution of 1:200 (1 hr, 37°C), and followed by goat anti-rabbit IgG Alexa Fluor 594 at a dilution of 1:600.
