- Western Blot: PKC theta [p Ser676] Antibody [NSB970] - Lysates prepared from Jurkat cells stimulated with PMA were resolved by SDS-PAGE on 10% polyacrylamide gel and transferred to PVDF. Membranes were left untreated (1-4) or treated with lambda (l) phosphatase (5), blocked with 5% BSA-TBST buffer ON at 4C, and incubated with 0.50mg/mL PKCq [pS676] Ab for 2hrs at RT in 3% BSA-TBST buffer, following prior incubation with: generic phosphoserine containing peptide (1), non-phosphopeptide corresponding to immunogen (2), phosphopeptide immunogen (3), or no peptide (4, 5). Data show that the peptide corresponding to PKCq [pS676] blocks the Ab signal, thereby demonstrating the specificity of the Ab; and that phosphatase stripping eliminates the signal, verifying that the Ab is phospho-specific.
