- Detection of Human CCL19/MIP-3 beta by Western Blot. Western blot shows conditioned media from monocyte-derived macrophages untreated (-) or treated (+) with 1 μg/mL LPS and 40 ng/mL Recombinant Human IFN-gamma (Catalog # 285-IF) for 24 hours. PVDF membrane was probed with 1 µg/mL of Goat Anti-Human CCL19/MIP-3 beta Antigen Affinity-purified Polyclonal Antibody (Catalog # AF361) followed by HRP-conjugated Anti-Goat IgG Secondary Antibody (Catalog # HAF017). A specific band was detected for CCL19/MIP-3 beta at approximately 12 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.

Simple Western lane view shows conditioned media from monocyte-derived macrophages untreated (-) or treated (+) with 1 μg/mL LPS and 40 ng/mL Recombinant Human IFN-gamma (Catalog # 285-IF) for 24 hours. A specific band was detected for CCL19/MIP-3 beta at approximately 8 kDa (as indicated) using 50 µg/mL of Goat Anti-Human CCL19/MIP-3 beta Antigen Affinity-purified Polyclonal Antibody (Catalog # AF361) followed by 1:50 dilution of HRP-conjugated Anti-Goat IgG Secondary Antibody (Catalog # HAF109). This experiment was conducted under reducing conditions and using the 12-230 kDa separation system.

- Chemotaxis Induced by CCL19/MIP-3 beta and Neutrali-zation by Human CCL19/MIP-3 beta Antibody. Re-combinant Human CCL19/MIP-3 beta (Catalog # 361-MI) chemoattracts the BaF3 mouse pro-B cell line transfected with human CCR7 in a dose-dependent manner (orange line). The amount of cells that migrated through to the lower chemotaxis chamber was measured by Resazurin (Catalog # AR002). Chemotaxis elicited by Recombinant Human CCL19/MIP-3 beta (50 ng/mL) is neutralized (green line) by increasing concentrations of Human CCL19/MIP-3 beta Antigen Affinity-purified Polyclonal Antibody (Catalog # AF361). The ND50 is typically 2-10 µg/mL.
