- Western blot was performed using Anti-SIRT6 Recombinant Rabbit Monoclonal Antibody (JJ20-69) (Product # MA5-32437) and a 40 kDa band corresponding to NAD-dependent protein deacetylase sirtuin-6 was observed across cell lines. Whole cell extracts (30 µg lysate) of HCT 116 (Lane 1), A549 (Lane 2), A-375 (Lane 3), SW480 (Lane 4), MCF7 (Lane 5), T-47D (Lane 6) were electrophoresed using NuPAGE™ 4-12% Bis-Tris Protein Gel (Product # NP0322BOX). Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1:1,000) and detected by chemiluminescence with Goat anti-Rabbit IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036,1:4000) using the iBright™ FL1500 Imaging System (Product # A44115). Chemiluminescentdetection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005)..

- Knockdown of NAD-dependent protein deacetylase sirtuin-6 was achieved by transfecting A549 with NAD-dependent protein deacetylase sirtuin-6 specific siRNAs (Silencer® select Product # S28307, S28306). Western blot analysis (Fig. a) was performed using Whole cell extracts from the NAD-dependent protein deacetylase sirtuin-6 knockdown cells (lane 3), non-targeting scrambled siRNA transfected cells (lane 2) and untransfected cells (lane 1). The blot was probed with SIRT6 Recombinant Rabbit Monoclonal Antibody (JJ20-69) (Product # MA5-32437, 1:1000 ) and Goat anti-Rabbit IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036, 1:4000). Densitometric analysis of this western blot is shown in histogram (Fig. b). Decrease in signal upon siRNA mediated knock down confirms that antibody is specific to NAD-dependent protein deacetylase sirtuin-6.
