- HL60 cells were left untreated (green) or treated with camptothecin to induce apoptosis (red). Cells were analyzed by intracellular flow cytometry using either the a D4 GDI monoclonal antibody (Product # MA1-41085) or a D4 GDI monoclonal antibody (Product # MA1-41181) (0.5 µg antibody/test/1 million cells). A: a D4 GDI monoclonal antibody (Product # MA1-41085) which recognizes only cleaved D4-GDI. B: a D4 GDI monoclonal antibody (Product # MA1-41085) which recognizes both full-length and cleaved D4-GDI. The results in (A) show that the untreated cell population was primarily negative for cleaved D4-GDI (Product # MA1-41085) whereas the treated population was primarily positive. This is because D4-GDI is cleaved in cells undergoing apoptosis but is present as a full-length form in normal cells. Hence the cleavage specific D4-GDI antibody (Product # MA1-41085) does not recognize very many cells in the untreated population. There may be a small percentage of positive cells in untreated populations due to a basal level of apoptosis. In contrast both the untreated and treated cell populations were positive for D4-GDI using Product # MA1-41181 (as seen in B). This is because it recognizes both the full-length (cells not undergoing apoptosis) and cleaved (apoptotic cells) forms of D4-GDI.
