- Western Blot using BAF57 Polyclonal Antibody (Product # PA5-28421). Various whole cell extracts (30 µg) were separated by 10% SDS-PAGE, and the membrane was blotted with BAF57 Polyclonal Antibody (Product # PA5-28421) diluted at 1:3,000. The HRP-conjugated anti-rabbit IgG antibody was used to detect the primary antibody.

- Knockdown of SWI/SNF-related matrix-associated actin-dependent regulator of chromatin subfamily E member 1 was achieved by transfecting HeLa cells with SWI/SNF-related matrix-associated actin-dependent regulator of chromatin subfamily E member 1 specific siRNAs (Silencer® select Product # S13160, S13162 ). An additional uncharacterized band (*) was observed along with the band of interest. Western blot analysis (Fig. a) was performed using Nuclear enriched extracts from the SWI/SNF-related matrix-associated actin-dependent regulator of chromatin subfamily E member 1 knockdown cells (lane 3), non-targeting scrambled siRNA transfected cells (lane 2) and untransfected cells (lane 1). The blot was probed with BAF57 Polyclonal Antibody (Product # PA5-28421, 1:2000 ) and Goat anti-Rabbit IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036, 1:4000). Densitometric analysis of this western blot is shown in histogram (Fig. b). A decrease in signal upon siRNA mediated knockdown confirms that antibody is specific to SWI/SNF-related matrix-associated actin-dependent regulator of chromatin subfamily E member 1.

- Western blot was performed using Anti-BAF57 Polyclonal Antibody (Product # PA5-28421) and a 57kDa band corresponding to SWI/SNF-related matrix-associated actin-dependent regulator of chromatin subfamily E member 1 was observed across cell lines and tissue tested. An additional non-specific band was observed in Hela cells. Nuclear enriched extracts (30 µg lysate) of HeLa (Lane 1), A-431 (Lane 2), Jurkat (Lane 3), MCF7 (Lane 4), Hep G2 (Lane 5), Raji (Lane 6), and Mouse Thymus (Lane 7) were electrophoresed using NuPAGE™ 4-12% Bis-Tris Protein Gel (Product # NP0322BOX). Resolved proteins were then transferred onto a Nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1:1000 dilution) and detected by chemiluminescence with Goat anti-Rabbit IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036, 1:4000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using Novex® ECL Reagent Kit (Product # WP20005).
