- Western blot was performed using Anti-E-cadherin Monoclonal Antibody (SHE78-7)(Product # 13-5700) and a 110kDa band corresponding to E-cadherin was observed across in all tested cell lines and tissues, except MDA-MB-231, SW480 and BJ. Whole cell extracts (30 µg lysate) of MCF7 (Lane 1), T-47D (Lane 2), MDA-MB-231 (Lane 3), SW480 (Lane 4), Caco-2 (Lane 5), BJ (Lane 6) were electrophoresed using NuPAGE™ 4-12% Bis-Tris Protein Gel (Product # NP0322BOX). Resolved proteins were then transferred onto a Nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (2 µg/mL) and detected by chemiluminescence with Goat anti-Mouse IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A28177,1:4000) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using SuperSignal™ West Dura Extended Duration Substrate (Product # 34076).The bands at ~80kDa and 60kDa corresponds to proteolytic cleavage products of E-cadherin.
