- Recombinant rat monoclonal antibodies are produced using in vitro expression systems. Recombinant antibodies are produced using specific genes that code for the desired antibodies. These genes are cloned into an expression vector and expressed in vitro. The advantages of recombinant antibodies include better specificity and lot-to-lot consistency. It is recommended that the antibody be carefully titrated for optimal performance in the assay of interest.

- Immunofluorescent analysis of IκB Zeta was performed using 70% confluent RAW 264.7 cells with or without LPS treatment (1 µg/mL, 6 hours). The cells were fixed with 4% paraformaldehyde for 10 minutes, permeabilized with 0.1% Triton X-100 for 15 minutes and blocked with 2% BSA for 1 hour at room temperature. The cells were stained with IκB Zeta Recombinant Rat Monoclonal Antibody (LK2NAP) (Product # 740065T, 1:200). Panels a) and e) shows representative images of cells that were stained for detection and localization of IκB Zeta. Panels b) and f) shows representative images of cells stained for nuclei using ProLong™ Diamond Antifade Mountant (Product # P36961) and Hoechst 33342, trihydrochloride trihydrate (Product #H1399, 1:300). Panels c) and g) shows representative images of cells stained for cytoskeletal F-actin (red) using Alexa Fluor™ 647 Phalloidin (Product # A22287, 1:300). Panels d) and h) are composite images of panels a), b), and c); and panels e), f), and g), respectively; clearly demonstrating nuclear translocation of IκB Zeta upon LPS treatment. The images were captured at 20X magnification and externally deconvoluted.

- RAW 264.7 cell line treated with LPS (1 µg/mL) for 6 hours (pink histogram) and without treatment (blue histogram) were fixed and permeabilized using the Foxp3 / Transcription Factor Staining Buffer Set (Product # 00-5523-00) and then stained intracellularly with 0.03 µg/test of IkB zeta Recombinant Rat Monoclonal Antibody (LK2NAP) (Product # 740065T), followed by Donkey anti-Rat IgG (H+L) Highly Cross-Adsorbed Secondary Antibody, Alexa Fluor™ 647 (Product # A78947). Viable cells were used for analysis as determined by Fixable Viability Dye eFluor™ 450 (Product # 65-0863-18). The flow cytometry data was acquired using Attune™ NxT Flow Cytometer (Product # A29004).
