- Western Blot Analysis/Peptide Blocking:Â K562 cell lysate was resolved by SDS-PAGE, transferred to PVDF, and probed with anti-phospho-CrkL (Tyr207) at a 1/1,000 dilution. Lane 1 (control) was not peptide blocked. Lanes 2 and 3 were peptide blocked with either non-phosphorylated peptide (Lane 2) or phosphorylated peptide (Lane 3) corresponding to the sequence surrounding Tyr207 of human CrkL. Proteins were visualized using a donkey anti-rabbit secondary antibody conjugated to HRP and a chemiluminescence detection system. Arrow indicates phosphorylated CrkL.
