Western Blot was performed using Anti-TMPRSS2 Polyclonal Antibody (Product # PA5-18847) and bands at 55, 28 kDa corresponding to full-length and cleaved TMPRSS2 were observed in LNCaP but not in DU 145 and PC-3 which express lower levels of TMPRSS2 (DOI: 10.1177/1535370214531880). Expression of both forms of TMPRSS2 increased upon treatment of LNCaP cells with DHT, no increase was observed in DU 145 and PC-3 (DOI: 10.1158/0008-5472.CAN-14-3297). Membrane enriched extracts (50 µg lysate) of LNCaP (Lane 1), DU 145 (Lane 2), PC-3 (Lane 3) (Fig. a); LNCaP (Lane 1), LNCaP treated with DHT (10 nm for 48 hours) (Lane 2), DU 145 (Lane 3), DU 145 treated with DHT (10 nm for 48 hours) (Lane 4), PC-3 (Lane 5), PC-3 treated with DHT (10 nm for 48 hours) (Lane 6) (Fig. b) were electrophoresed using NuPAGE™ 4-12% Bis-Tris Protein Gel (Product # NP0321BOX). Resolved proteins were then transferred onto a Nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The Blot was probed with the primary antibody (0.5 µg/mL dilution) and detected by chemiluminescence with Rabbit anti-Goat IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27014, 1:4000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005). Two uncharacterized bands (*) were observed at ~70, 80 kDa.
