- Immunofluorescent analysis of Oct4/Pou5F1 (green) shows nuclear staining in human HEL 11.4 iPS cells, indicating pluripotency. Oct4/Pou5F1 staining (left panel), DAPI staining (middle panel) and an overlay image of Oct4/Pou5F1 with DAPI (right panel) are shown. HEL 11.4 iPS cells were grown on matrigel-coated chamber slides and fixed with 4% formaldehyde for 15 minutes prior to staining. Cells were probed with a DyLight 488-conjugated Oct4/Pou5F1 monoclonal antibody (Product # MA1-104-D488) in 0.1% Tween-PBS at a dilution of 1:20, and incubated overnight at 4C. Images were taken at 20X magnification.

- Immunofluorescence analysis of OCT-4 was performed using 70% confluent log phase NTERA-2 cells. The cells were fixed with 4% paraformaldehyde for 10 minutes, permeabilized with 0.1% Triton™ X-100 for 15 minutes, and blocked with 1% BSA for 1 hour at room temperature. The cells were labeled with OCT-4 Mouse Monoclonal Antibody (Product # MA1-104-D488) at 5 µg/mL in 0.1% BSA, incubated at 4 degree celsius overnight (Panel a: green). Nuclei (Panel b: blue) were stained with ProLong™ Diamond Antifade Mountant with DAPI (Product # P36962). F-actin (Panel c: red) was stained with Rhodamine Phalloidin (Product # R415, 1:300). Panel d represents the merged image showing nuclear localization. Panel e shows OCT-4 negative cell line HeLa with no signal. Panel f represents control cells with Isotype control to assess background. The images were captured at 60X magnification.
