- CD4-enriched human peripheral blood cells were polarized under Th1 conditions (with Human IL-12 Recombinant Protein (Product # 34-8129-82) and Human IL-2 Recombinant Protein (Product # BMS334) for 10 days. Cells were restimulated with Protein Transport Inhibitor Cocktail (Product #00-4980-03) (left) or with Cell Stimulation Cocktail plus Protein Transport Inhibitors (Product #00-4975-03) (right) for 5 hours. Cells were then stained intracellularly, using the Intracellular Fixation & Permeabilization Buffer Set (Product # 88-8824-00) and protocol, with CD4 Monoclonal Antibody, FITC (Product # 11-0049-42) and 0.125 µg TNF beta Monoclonal Antibody, APC. Cells in the lymphocyte gate were used for analysis.
