Pateclizumab (also known as MLTA-3698A or RG-7418) is an investigational, humanized monoclonal antibody that targets lymphotoxin-alpha (LT¦Á, also known as TNF-¦Â), a cytokine belonging to the tumor necrosis factor (TNF) superfamily. LT¦Á exists both as a soluble homotrimer and as a membrane-bound heterotrimer with lymphotoxin-beta (LT¦Â), which signals through the LT¦Â receptor. By binding to LT¦Á, pateclizumab neutralizes its activity, thereby inhibiting signaling through both TNF receptors and the LT¦Â receptor. This blockade aims to disrupt the formation and maintenance of lymphoid-like structures in inflamed tissues, which are thought to drive chronic inflammation in autoimmune diseases. It has been clinically evaluated for the treatment of rheumatoid arthritis (RA).
