- Knockdown of SRCAP was achieved by transfecting SK-MEL-147 with SRCAP specific shRNAs. Western blot analysis was performed on NuPAGE™ 3-8% Tris-Acetate Protein Gel (Product # EA0378BOX) using chromatin lysate (purified from SK-MEL-147) from SRCAP knockdown cells (shSRCAP #2, lane 3), SRCAP knockdown cells (shSRCAP #1, lane 2) and untransfected cells (lane 1). The blot was probed with SRCAP Polyclonal Antibody (Product # PA5-56012, 1:500 dilution) and anti-Rabbit Secondary Antibody, HRP conjugate. Decrease in band intensity upon shRNA mediated knock down confirms that antibody is specific to SRCAP.
