Experimental details

Detection of Phospho-GABA_B R2 (S892) by Western Blot Western blots of COS-7 cells transiently expressing mutant GABA_B R2 (S892A) (lane 1), wild type GABA_B R2 (lane 2) or control untransfected cells (lane 3) were lysed and 50 mg of protein from each sample was separated by SDS-PAGE and transferred to a nitrocellulose membrane. Blots were then immunolabeled with rabbit anti-phospho-GABA_B R2 (S892) (A). In B, the membrane was pre-treated with lambda-PPase (1200 units for 30 minutes) before being exposed to the anti-GABA_B Receptor, R2 subunit (S892). The immunolabeling is completely eliminated by treatment with lambda-PPase.
