- Western blot analysis was performed on whole cell extracts (30 µg lysate) of HT-29 (Lane 1), SW-480 (Lane 2), Jurkat (Lane 3), U-2OS (Lane 4), NIH/3T3 (Lane 5), HEK-293 (Lane 6), MDA-MB-231 (Lane 7) and HCT 116 (Lane 8). The blot was probed with Anti-AXIN2 Polyclonal Antibody (Product # PA5-25331, 1:1000 dilution) and detected by chemiluminescence using Goat anti-Rabbit IgG (H+L) Superclonal™ Secondary Antibody, HRP conjugate (Product # A27036, 0.25 µg/ml, 1:4000 dilution). A 94 kDa band corresponding to AXIN2 was observed across the cell lines tested.

- Western blot analysis of Axin2 was performed by loading 30 µg of HT29 Cas9 control (Lane 1) and HT29 beta catenin KO whole cell lysate. The blot was probed with AXIN2 Polyclonal Antibody (Product # PA5-25331, 1:1000 dilution) and Goat anti-Rabbit IgG (H+L) Superclonal™ Secondary Antibody, HRP conjugate (Product # A27036, 0.25 µg/ml, 1:4000 dilution. Loss of signal upon CRISPR mediated knockout (KO) confirms that antibody is specific to Axin2. A non-specific band was also observed at ~35 kDa with the above testing conditions.
