Description

Lumretuzumab (also known as RG-7116) is a humanized, glycoengineered monoclonal antibody that targets HER3 (ErbB3), a member of the epidermal growth factor receptor (EGFR/ErbB) family. The glycoengineering of its Fc region enhances antibody-dependent cellular cytotoxicity (ADCC). By binding to HER3, lumretuzumab blocks the receptor's ability to dimerize with other ErbB family members (particularly HER2), thereby inhibiting downstream oncogenic signaling pathways such as PI3K/AKT. It was developed as a potential therapeutic agent for HER3-expressing solid tumors, with clinical trials focusing on breast cancer, gastric cancer, and non-small cell lung cancer (NSCLC), often in combination with other agents like pertuzumab (anti-HER2) or chemotherapy.
