- Description
- Activation of NF°ÙB requires that I°ÙB be phosphorylated on specific serine residues, which results in targeted degradation of I°ÙB. I°ÙB kinase Ï« (IKKÏ«), previously designated CHUK, interacts with I°ÙB-Ï« and specifically phosphorylates I°ÙB-Ï« on the sites that trigger its degradation Serines 32 and 36. IKKÏ« appears to be critical for NF°ÙB activation in response to proinflammatory cytokines. Phosphorylation of I°ÙB by IKKÏ« is stimulated by the NF°ÙB inducing kinase (NIK), which itself is a central regulator for NF°ÙB activation in response to TNF and IL-1. The functional IKK complex contains three subunits, IKKÏ«, IKKÉÇ and IKKÎÛ , and each appear to make essential contributions to I°ÙB phosphorylation.
