- Extracts of A431 cells treated with with 200 ng/mL EGF for 15 minutes were resolved by SDS-PAGE on a 10% Tris-glycine gel and transferred to PVDF. The membranes were left untreated (1-4, 6) or treated with lambda phosphatase (5), blocked with a 4% BSA-TBST buffer for one hour at room temperature, and incubated with the MEK2 [pT394] (human) antibody for two hours at room temperature in a 1% BSA-TBST, following prior incubation with: no peptide (1, 5, 6), the non-phosphopeptide corresponding to the phosphoepptide immunogen (2), a generic phosphothreonine-containing peptide (3) or the phosphopeptide immunogen (4). After washing, the membranes were incubated with goat F(ab)2 anti-rabbit IgG: HRP conjμgate and signals were detected. The data show that only the phosphopeptide corresponding to MEK2 [pT394] (human) blocks the antibody signal. The data also show that phosphatase stripping eliminates the signal, further verifying that the antibody is phospho-specific.
