Description

mTOR (mammalian target of rapamycin) is a Ser/Thr kinase acting as an ATP sensor and is reported to autophosphorylate Ser2481 in the presence of sufficient nutrients. Activated mTOR phosphorylates ribosomal protein S6 kinase 1 (S6K1) and the eukaryotic translation initiation factor 4E binding protein 1 (4EBP1) in response to PI3K/Akt signaling, in turn, promoting cell growth, survival, and proliferation.

Experimental details

Myc-mTOR transfected 293 cells were starved for 30 hours, then treated with (lane 1) or without (lane 2) 20% FBS for 30 minutes. The cell lysates were resolved by electrophoresis, transferred to nitrocellulose, and probed with the Poly 6517 anti-mTOR pS2481 antibody. Proteins were visualized using a donkey anti-rabbit-IgG secondary conjugated to HRP and chemiluminescence detection.
