Experimental details: Western blot was performed using Anti-TRAIL Monoclonal Antibody (55B709.3) (Product # MA1-41027) and 28, 30 kDa bands corresponding to Tumor necrosis factor ligand superfamily member 10 were observed. Whole cell extracts (30 µg lysate) of A-431 (Lane 1), A-431 treated with Protein Transport Inhibitor (PTI) cocktail (1X, 4 hours) (Lane 2), HaCaT (Lane 3), HaCaT treated with Protein Transport Inhibitor (PTI) cocktail (1X, 4 hours) (Lane 4), BeWo (Lane 5), HT-29 (Lane 6), HT-29 treated with Protein Transport Inhibitor (PTI) cocktail (1X, 4 hours) (Lane 7), U-2 OS (Lane 8) and U-2 OS treated with Protein Transport Inhibitor (PTI) cocktail (1X, 4 hours) (Lane 9) were electrophoresed using NuPAGE™ 10% Bis-Tris Protein Gel (Product # NP0302BOX). Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1.5 µg/mL) and detected by chemiluminescence with Goat anti-Mouse IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A28177,1:20,000) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using SuperSignal™ West Pico PLUS Chemiluminescent Substrate (Product # 34580). Bands of interest were not observed in HT-29 and U-2 OS as expected.
