- Knockdown of LAMTOR3 was achieved by transfecting HeLa with LAMTOR3 specific siRNAs (Silencer® select Product # s16462, s16464). Western blot analysis (Fig. a) was performed using whole cell extracts from the LAMTOR3 knockdown cells (lane 3), non-specific scrambled siRNA transfected cells (lane 2) and untransfected cells (lane 1). The blots were probed with LAMTOR3 Polyclonal Antibody (Product # PA5-22145, 1:1000 dilution) and Goat anti-Rabbit IgG (H+L) Superclonal™ Secondary Antibody, HRP conjugate (Product # A27036, 0.25µg/mL, 1:4000 dilution). Densitometric analysis of this western blot is shown in histogram (Fig. b). Decrease in signal upon siRNA mediated knock down confirms that antibody is specific to LAMTOR3.

- Western blot analysis was performed on whole cell extracts (30 µg lysate) of HeLa (Lane 1), PANC-1 (Lane 2), MOLT-4 (Lane 3), K-562 (Lane 4), C2C12 (Lane 5), RAW 264.7 (Lane 6), Mouse Brain (Lane 7), Mouse Lung (Lane 8) and Mouse Testis (Lane 9). The blot was probed with Anti-LAMTOR3 Polyclonal Antibody (Product # PA5-22145, 1:1000 dilution) and detected by chemiluminescence Goat anti-Rabbit IgG (H+L) Superclonal™ Secondary Antibody, HRP conjugate (Product # A27036, 0.25 µg/mL, 1:4000 dilution). A 13kDa band corresponding to LAMTOR3 was observed across all the cell lines and tissues tested. Some uncharacterized bands were observed in case of mouse tissue lysates.
