Experimental details: Knockdown of LAMTOR3 was achieved by transfecting HeLa with LAMTOR3 specific siRNAs (Silencer® select Product # s16462, s16464). Western blot analysis (Fig. a) was performed using whole cell extracts from the LAMTOR3 knockdown cells (lane 3), non-specific scrambled siRNA transfected cells (lane 2) and untransfected cells (lane 1). The blots were probed with LAMTOR3 Polyclonal Antibody (Product # PA5-22145, 1:1000 dilution) and Goat anti-Rabbit IgG (H+L) Superclonal™ Secondary Antibody, HRP conjugate (Product # A27036, 0.25µg/mL, 1:4000 dilution). Densitometric analysis of this western blot is shown in histogram (Fig. b). Decrease in signal upon siRNA mediated knock down confirms that antibody is specific to LAMTOR3.

Experimental details: Western blot analysis was performed on whole cell extracts (30 µg lysate) of HeLa (Lane 1), PANC-1 (Lane 2), MOLT-4 (Lane 3), K-562 (Lane 4), C2C12 (Lane 5), RAW 264.7 (Lane 6), Mouse Brain (Lane 7), Mouse Lung (Lane 8) and Mouse Testis (Lane 9). The blot was probed with Anti-LAMTOR3 Polyclonal Antibody (Product # PA5-22145, 1:1000 dilution) and detected by chemiluminescence Goat anti-Rabbit IgG (H+L) Superclonal™ Secondary Antibody, HRP conjugate (Product # A27036, 0.25 µg/mL, 1:4000 dilution). A 13kDa band corresponding to LAMTOR3 was observed across all the cell lines and tissues tested. Some uncharacterized bands were observed in case of mouse tissue lysates.
