Experimental details

Western blot analysis from lysates of human brain tissue and mouse brain tissue. PVDF membrane was probed with 1 µg/mL Sheep Anti-human/mouse PLC-ß1 Antigen Affinity-purified Polyclonal Antibody (Product # PA5-48052) followed by HRP-conjugated Anti-Sheep IgG Secondary Antibody. A specific band for PLC-ß1 was detected at approximately 150 kDa (as indicated). This experiment was conducted under reducing conditions.

Western blot analysis of PLCB1 in 5 ng/lane recombinant human PLC-β1, PLC-β2, PLC-β3, PLC-β4. Samples were incubated in PLCB1 polyclonal antibody (Product # PA5-48052) using a dilution of 1 µg/mL followed by a HRP-conjugated Anti-Sheep IgG secondary antibody. A specific band for PLC‚β1 was detected at approximately 28 kDa (as indicated). This experiment was conducted under reducing conditions.

Western blot was performed using Anti-PLCB1 Polyclonal Antibody (Product # PA5-48052) and a 160 kDa band corresponding to PLCB1 was observed along with uncharacterized bands. Tissue extracts (30 µg lysate) of Mouse Brain (Lane 1), Mouse Cerebellum (Lane 2), Mouse Heart (Lane 3), Rat Brain (Lane 4) and Rat Heart (Lane 5) were electrophoresed using NuPAGE™ 4-12% Bis-Tris Protein Gel (Product # NP0322BOX). Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1 µg/mL) and detected by chemiluminescence with Rabbit anti-Sheep IgG (H+L) Secondary Antibody, HRP (Product # 61-8620, 1:4000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using SuperSignal™ West Dura Extended Duration Substrate (Product # 34076).
