- Immunoblot Analysis: Lysates from unstimulated (lanes 1,3) and EGFstimulated (lanes 2,4) A431 cells were resolved by electrophoresis, transferred to nitrocellulose and probed with anti-phospho-PLCγ-1 (Tyr771)(2μg/ml). Prior to electrophoresis, samples in lanes 3 and 4 were treated with λ-phosphatase for 1 hour at 30°C. Proteins were visualized using a Goat anti-Rabbit secondary antibody conjugated to HRP and a chemiluminescence detection system. Arrow indicates phospho-PLC γ-1 (~135kDa).
