Description

STAT5 has gene transactivation activity and plays a key role in lymphocyte differentiation and proliferation. Tyrosine phosphorylation of STAT5, induced by cytokine stimulation, results in dimerization through its SH2 domains. The dimerized STAT5 proteins translocates from the cytoplasm to the nucleus and subsequently regulates gene transcription.

Experimental details

U937 whole cell extracts were resolved by electrophoresis, transferred to nitrocellulose, and probed with polyclonal anti-STAT 5 pY694 (Poly6511) antibody. Proteins were visualized using anti-rabbit IgG secondary antibody conjugated to HRP and chemiluminescence detection. Lane 1 - lysate from untreated U937 cells treated with 10 ng/ml human GM-CSF for 30 minutes. The same blot was stripped and re-probed with β-actin antibody (Poly6221) to ensure equal amounts of lysates were loaded in both lanes.
