- Detection of Recombinant Mouse and Rat IL-1 alpha/IL-1F1 by Western Blot. Western blot shows 50 ng of Recombinant Mouse IL-1 alpha/IL-1F1 (Catalog # 400-ML), Recombinant Human IL-1 alpha/IL-1F1 (Catalog # 200-LA) and Recombinant Rat IL-1 alpha/IL-1F1 (Catalog # 500-RL). PVDF Membrane was probed with 0.1 µg/mL of Goat Anti-Mouse IL-1 alpha/IL-1F1 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF-400-NA) followed by HRP-conjugated Anti-Goat IgG Secondary Antibody (Catalog # HAF109). A specific band was detected for IL-1 alpha/IL-1F1 at approximately 16 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 3.

IL-1 alpha/IL-1F1 was detected in perfusion fixed frozen sections of mouse liver using Goat Anti-Mouse IL-1 alpha/IL-1F1 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF-400-NA) at 15 µg/mL overnight at 4 °C. Tissue was stained using the Anti-Goat HRP-DAB Cell & Tissue Staining Kit (brown; Catalog # CTS008) and counterstained with hematoxylin (blue). Recombinant Mouse IL-1 alpha/IL-1F1 (Catalog # 400-ML) stimulates proliferation in the the D10.G4.1 mouse helper T cell line in a dose-dependent manner (orange line). Proliferation elicited by Recombinant Mouse IL-1 alpha/IL-1F1 (50 pg/mL) is neutralized (green line) by increasing concentrations of Goat Anti-Mouse IL-1 alpha/IL-1F1 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF-400-NA). The ND50 is typically 0.002-0.004 µg/mL in the presence of concanavalin A (1.25 µg/mL).
