Western blot was performed using Anti-VDR Polyclonal Antibody (Product # PA1-9116) and a 60kDa band corresponding to VDR was observed in all the tested cell models, and was upregulated upon LPS treatment in RAW 264.7. Whole cell lysate (30ug lysate) of HeLa (Lane 1), LNCaP (Lane 2), C2C12 (Lane 3), SK-OV-3 (Lane 4), RAW264.7 (Lane 5), RAW264.7 (LPS, 10µg/ml, 24hrs) (Lane 6) and THP-1 (Lane 7) were electrophoresed using Novex® NuPAGE® 4-12 % Bis-Tris gel (Product # NP0322BOX). Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1µg/ml concentration) and detected by chemiluminescence with Rabbit anti-Goat IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27014, 1:4000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using SuperSignal™ West Dura Extended Duration Substrate (Product # 34076).
