- Knockdown of SHP2 was achieved by transfecting A-431 cells with SHP2 specific siRNAs (Silencer® select Product # s11526). Western blot analysis (Fig. a) was performed using whole cell extracts SHP2 knockdown cells (lane 3), non-specific scrambled siRNA transfected cells (lane 2) and untransfected cells (lane 1). The blots were probed with SHP2 Polyclonal Antibody (Product # PA5-17956, 1:1000 dilution) and Rabbit anti-Goat IgG (H+L) Superclonal™ Secondary Antibody, HRP conjugate (Product # A27014, 1 mg/mL, 1:4000 dilution). Densitometric analysis of this western blot is shown in histogram (Fig. b). Decrease in signal upon siRNA mediated knock down confirms that antibody is specific to SHP2.

- Western blot analysis was performed on whole cell extracts (30 µg lysate) of HT-29 (Lane 1), HeLa (Lane 2), C2C12 (Lane 3) and HEK 293 (Lane 4). The blot was probed with Anti-SHP2 Polyclonal Antibody (Product # PA5-17956, 1 µg/mL) and detected by chemiluminescence using Goat anti-Goat IgG (H+L) Superclonal™ Secondary Antibody, HRP conjugate (Product # A27014, 0.25 µg/mL, 1:4000 dilution). A 72 kDa band corresponding to SHP2 was observed across the cell lines tested.

- Flow cytometric analysis of SHP2 in A431 cells using a polyclonal antibody (Product #PA5-17956). A431 cells (blue line) were paraformaldehyde fixed and permeabilized with 0.5% Triton. The primary antibody was incubated for one hour (10 µg/mL) followed by an Alexa Fluor 488 secondary antibody (1 µg/mL). IgG control: Unimmunized goat IgG (black line) followed by an Alexa Fluor 488 secondary antibody.
