Experimental details

Western blot was performed using Anti-TIMP2 Polyclonal Antibody (Product # PA1-25313) and a 22kDa band corresponding to TIMP2 was observed across cell lines tested except for Daudi which is reported to be negative. Whole cell extracts (30 µg lysate) of SH-SY5Y (Lane 1), SH-SY5Y treated with Protein Transport Inhibitor (1x for 4 hours) (Lane 2), HT-1080 (Lane 3), HT-1080 treated with Protein Transport Inhibitor (1x for 4 hours) (Lane 4), Daudi (Lane 5) and Daudi treated with Protein Transport Inhibitor (1x for 4 hours) were electrophoresed using Novex® NuPAGE® 4-12 % Bis-Tris gel (Product # NP0322BOX). Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1:1000 dilution) and detected by chemiluminescence with Goat anti-Rabbit IgG (H+L), Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036, 1:4000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005). Two uncharacterized bands (*) were observed at ~50 and ~110kDa respectively.
