- Western blot analysis was performed on tissue extracts (30 µg lysate) of Mouse colon (Lane 1), and Mouse spleen (Lane 2). The blots were probed with Anti-TIMP2 Mouse Monoclonal Antibody (Product # MA5-13603, 1-2 µg/mL) and detected by chemiluminescence using Goat anti-Mouse IgG (H+L) Superclonal™ Secondary Antibody, HRP conjugate (Product # A28177, 0.4 µg/mL, 1:2500 dilution). A ~ 24 kDa band corresponding to TIMP2 was observed across tissues tested. Known quantity of protein samples were electrophoresed using Novex® NuPAGE® 4-12 % Bis-Tris gel (Product # NP0321BOX), XCell SureLock™ Electrophoresis System (Product # EI0002) and Novex® Sharp Pre-Stained Protein Standard (Product # LC5800). Resolved proteins were then transferred onto a nitrocellulose membrane with iBlot® 2 Dry Blotting System (Product # IB21001). The membrane was probed with the relevant primary and secondary Antibody following blocking with 5 % skimmed milk. Chemiluminescent detection was performed using Pierce™ ECL Western Blotting Substrate (Product # 32106).

- Western blot was performed using Anti-TIMP2 Monoclonal Antibody (T2-101) (Product # MA5-13603) and a 24kDa band corresponding to TIMP2 was observed in HT-1080 but was absent in Daudi which is reported to be negative. Whole cell extracts (30 µg lysate) of HT-1080 (Lane 1), HT-1080 treated with Protein Transport Inhibitor (PTI; 1x for 4 hours) (Lane 2), Daudi (Lane 3) and Daudi treated with Protein Transport Inhibitor (1x for 4 hours) (Lane 4) were electrophoresed using Novex® NuPAGE® 4-12 % Bis-Tris gel (Product # NP0322BOX). Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1µg/mL) and detected by chemiluminescence with Goat anti-Mouse IgG (H+L), Superclonal™ Recombinant Secondary Antibody, HRP (Product # A28177, 1:4000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005).
