- Western Blot analysis of LIFR in extracts of mouse skeletal muscle using LIFR Polyclonal Antibody (Product # PA5-88754) at a dilution of 1:1000. A HRP Goat Anti-Rabbit IgG (H+L) secondary antibody was used at a dilution of 1:10,000. Lysates/proteins: 25 µg per lane. Blocking buffer: 3% nonfat dry milk in TBST.

Samples were incubated with LIFR Polyclonal antibody (Product # PA5-88754) using a dilution of 1:3,000, followed by HRP Goat Anti-Rabbit IgG (H+L) at a dilution of 1:10,000. Lysates/proteins: 25 µg per lane. Blocking buffer: 3% nonfat dry milk in TBST. Detection: ECL Enhanced Kit. Exposure time: 30s.

- Western blot was performed using LIFR Polyclonal Antibody (Product # PA5-88754) and a 190 kDa band corresponding to LIFR was observed across all cell lines tested except for SW480 and HCT 116, in which, LIFR promoter methylation has been reported. Non-specific bands (*) were also observed around ~90 kDa and 35 kDa.Whole-cell extracts (40 µg lysate) of HeLa (Lane 1), SiHa (Lane 2), SW480 (Lane 3), RD (Lane 4), and HCT 116 (Lane 5) were electrophoresed using NuPAGE™ 3-8% Tris-Acetate Protein Gel (Product # EA0378BOX), 10 well. Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1:1000 dilution) and detected by chemiluminescence with Goat anti-Rabbit IgG (Heavy Chain) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036, 1:20,000 dilution) using the iBright™ FL1500 Imaging System (Product # A44115). Chemiluminescent detection was performed using SuperSignal™ West Pico PLUS Chemiluminescent Substrate (Product # 34580).
