- Western blot analysis was performed on whole cell extracts (30 µg lysate) of A549 (Lane 1) and A549 treated with SB216763 (10uM for 30 min) followed by EGF (200 ng/mL for 10 min) (Lane 2). The blot was probed with Anti-Phospho-GSK3 alpha/beta (Ser9, Ser21) Rabbit Monoclonal Antibody (Product # MA5-15023, 1:500 dilution) and detected by chemiluminescence using Goat anti-Rabbit IgG (H+L) Superclonal™ Secondary Antibody, HRP conjugate (Product # A27036, 0.25 µg/mL, 1:4000 dilution). A 51 kDa band corresponding to Phospho-GSK3 alpha was detected in untreated lysate and decreased upon SB216763 treatment.
