- Western blot analysis was performed on membrane enriched extracts (30 µg lysate) of MDA-MB-231 (Lane 1), PANC1 (Lane 2) and SKOV3 (Lane 3). The blots were probed with Mouse Anti-CD171 Monoclonal Antibody, Biotin (Product # MA5-14137, 1 µg/mL) and detected by chemiluminescence using Poly-HRP Streptavidin (Product # N200, 1:10,000 dilution). A 200 kDa corresponding to CD171 major form along with lower molecular mass form of 80 kDa corresponding to intracellular region of protein was observed across the cell lines tested. Known quantity of protein samples were electrophoresed using Novex® NuPAGE® 4-12 % Bis-Tris gel (Product # NP0322BOX), XCell SureLock™ Electrophoresis System (Product # EI0002) and Novex® Sharp Pre-Stained Protein Standard (Product # LC5800). Resolved proteins were then transferred onto a nitrocellulose membrane with wet transfer system. The membrane was probed with the relevant primary antibody following blocking with 5 % skimmed milk. Chemiluminescent detection was performed using Pierce™ ECL Western Blotting Substrate (Product # 32106).

- Knockout of CD171 was achieved by CRISPR-Cas9 genome editing. Western blot analysis of CD171 was performed by loading 20 µg of HeLa wild type (Lane 1), HeLa Cas9 control (Lane 2), HeLa CD171 knockout (Lane 3) membrane enriched cell extracts. The blot was probed with Anti-CD171Monoclonal Antibody (UJ127), Biotin(Product # MA5-14137) using 1 µg/mL and Goat anti-Mouse IgG (H+L), Superclonal™ Recombinant Secondary Antibody, HRP (Product # A28177) using 1:4000 dilution. Loss of signal upon CRISPR mediated knockout (KO) confirms that antibody is specific to CD171.

- Western blot was performed using Anti-CD171 Monoclonal Antibody (UJ127), Biotin(Product # MA5-14137) and a 250, 85kDa band corresponding to CD171 was observed across the panel tested except Caco-2 which is reported to be negative. Whole cell extracts (30 µg lysate) of SW480 (Lane 1), Caco-2 (Lane 2), PANC-1 (Lane 3) and U-2 OS (Lane 4) were electrophoresed using NuPAGE™ 4-12% Bis-Tris Protein Gel (Product # NP0321BOX). Resolved proteins were then transferred onto a Nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1ug/ml) and detected by chemiluminescence with Poly- HRP Streptavidin (Product # N200,1:4000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005).
