- Western Blot was performed using Anti-Phospho-STAT4 (Ser721) Polyclonal Antibody (Product # PA5-105861) and a 86 kDa band corresponding to Signal transducer and activator of transcription 4 was observed across cell lines tested . Whole cell extracts (50 µg lysate) of NK-92 untreated (Lane 1), NK-92 treated with Hu IL2 (Lane 2), NK-92 untreated (Lane 3) and NK-92 treated with Hu IL 12 (Lane 4) were electrophoresed using NuPAGE™ 4-12% Bis-Tris Protein Gel (Product # NP0321BOX). Resolved proteins were then transferred onto a nitrocellulose membrane (Product # LC2001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1:1000) and detected by chemiluminescence with Goat anti-Rabbit IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036, 1:4000) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005).Expression of Phospho STAT-4 was significantly upregulated upon treatment with IL-2 and IL-12.

- Immunofluorescent analysis of Phospho-STAT4 (Ser721) in K562. Samples were fixed with paraformaldehyde, permeabilized with 0.1% Triton X-100, blocked with 10% serum (45 min at 25°C) incubated with Phospho-STAT4 (Ser721) polyclonal antibody (Product # PA5-105861) using a dilution of 1:200 (1 hr, 37°C), and followed by goat anti-rabbit IgG Alexa Fluor 594 at a dilution of 1:600.
