- Western blot was performed using Anti-VAV1 Monoclonal Antibody (2E11) (Product # MA5-17198) and a 98kDa band corresponding to VAV1 was observed in Raji, MOLT-4, Jurkat, THP-1 but not in T-47D and MCF7 which are reported to be negative. Whole cell extracts (30ug) of Raji (Lane 1), MOLT-4 (Lane 2), Jurkat (Lane 3), THP-1 (Lane 4), T-47D (Lane 5) and MCF7 (Lane 6) were electrophoresed using NuPAGE® 10 % Bis-Tris gel (Product # NP0302BOX). Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1:1000 dilution) and detected by chemiluminescence with Goat anti-Mouse IgG (H+L), Superclonal™ Recombinant Secondary Antibody, HRP (Product # A28177, 1:4000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using SuperSignal™ West Dura Extended Duration Substrate (Product # 34076).
