Western blot was performed using Anti-Estrogen Receptor alpha Monoclonal Antibody (6F11) (Product # MA1-27107) and a 66 kDa band corresponding to ESR1 was observed across tissues and cell lines tested. Membrane enriched extracts (30 µg lysate) of Mouse Uterus (Lane 1), Rat Uterus (Lane 2), Mouse Ovary (Lane 3), Rat Ovary (Lane 4), Mouse Lung (Lane 5), Rat Lung (Lane 6), Mouse Brain (Lane 7), T-47D (Lane 8), SK-BR-3 (Lane 9) were electrophoresed using NuPAGE™ 4-12% Bis-Tris Protein Gel (Product # NP0322BOX). Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1:500 dilution) and detected by chemiluminescence with Goat anti-Mouse IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A28177,1:4000) using the iBright™ FL1500 Imaging System (Product # A44115). Experimental details

Immunofluorescence analysis of Estrogen Receptor alpha was performed using 70% confluent log phase T-47D cells. The cells were fixed with 4% paraformaldehyde for 10 minutes, permeabilized with 0.1% Triton™ X-100 for 15 minutes, and blocked with 2% BSA for 45 minutes at room temperature. The cells were labeled with beta Estrogen Receptor alpha Monoclonal Antibody (6F11) (Product # MA1-27107) at 1:200 dilution in 0.1% BSA, incubated at 4 degree celsius overnight and then labeled with Goat anti-Mouse IgG (H+L) Highly Cross-Adsorbed Secondary Antibody, Alexa Fluor Plus 488 (Product # A32723), (1:2000 dilution), for 45 minutes at room temperature (Panel a: Green). Nuclei (Panel b: Blue) were stained with ProLong™ Diamond Antifade Mountant with DAPI (Product # P36962). F-actin (Panel c: Red) was stained with Rhodamine Phalloidin (Product # R415, 1:300). Panel d represents the merged image showing nuclear localization. Panel e represents merged image for SK-BR-3 cells showing no staining for Estrogen Receptor alpha. Panel f represents control cells with no primary antibody to assess background. The images were captured at 60X magnification.

Chromatin Immunoprecipitation (ChIP) assay of endogenous Estrogen Receptor alpha protein using ESR alpha Antibody: ChIP was performed using Anti-Estrogen Receptor alpha Monoclonal Antibody (6F11) (Product # MA1-27107, 5 ul) on sheared chromatin from MCF7 cells treated with beta estradiol (10 nm for 45 mins) using the MAGnify ChIP System kit (Product # 49-2024). Chromatin from untreated MCF7 cells were prepared the same way. Normal Mouse IgG was used as a negative IP control. The purified DNA was analyzed by qPCR using primers binding to XBP1, GREB1 (Active) and GAPDH (+4.5kb downstream of promoter) and SAT2 satellite repeats (Inactive). Data is presented as fold enrichment of the antibody signal versus the negative control IgG using the comparative CT method.
