- Western Blot analysis of CBS was performed by separating 30 µg of non-transfected (–) and transfected (+) 293T whole cell extracts by 5% SDS-PAGE. Proteins were transferred to a membrane and probed with a CBS Monoclonal Antibody (GT519) (Product # MA5-17273) at a dilution of 1:1000. The HRP-conjugated anti-mouse IgG antibody was used to detect the primary antibody.

- Western blot was performed using Anti-CBS Monoclonal Antibody (GT519) (Product # MA5-17273) and a band at 61 kDa corresponding to CBS was observed across the cell lines and tissues tested. OVCAR-3 was observed to be high expressing cell line when compared to SK-OV-3; Mouse, Rat Liver and Mouse Pancreas were observed to be high expressing tissues when compared to Mouse and Rat Heart as reported in the literature. Whole cell extracts (30 µg lysate) of Hep G2 (Lane 1), HEK-293 (Lane 2), HeLa (Lane 3), OVCAR-3 (Lane 4), SK-OV-3 (Lane 5), DU 145 (Lane 6), tissue extracts of Mouse Liver (Lane 7), Rat Liver (Lane 8), Mouse Pancreas (Lane 9), Mouse Heart (Lane 10) and Rat Heart (Lane 11) were electrophoresed using Novex® NuPAGE® 4-12% % Bis-Tris gel (Product # NP0322BOX). Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1:1000 dilution) and detected by chemiluminescence with Goat anti-Mouse IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A28177, 1:4000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005).
