Description
Gevokizumab (development code XOMA-052) is an investigational, humanized monoclonal IgG2 antibody developed for the treatment of auto-inflammatory diseases. It represents a unique approach within the class of interleukin-1 beta (IL-1¦Â) inhibitors. Gevokizumab functions as an allosteric modulator of the IL-1¦Â pathway. Unlike direct neutralizing antibodies, it binds to a specific, allosteric site on the IL-1¦Â molecule itself. This binding reduces the affinity of IL-1¦Â for its type I receptor (IL-1RI) without completely blocking the interaction. By modulating rather than completely inhibiting the signal, gevokizumab aims to dampen the excessive inflammatory response driven by IL-1¦Â while potentially preserving some of its physiological functions, thereby offering a targeted and potentially safer anti-inflammatory effect.
