- Immunofluorescent analysis of ZO-1 (red) in Caco-2 cells. The cells were fixed with 4% Paraformaldehyde in PBS for 15 minutes at room temperature, and blocked with 3% BSA in PBS (Product # 37525) for 30 minutes at room temperature. Cells were stained with a ZO-1 Monoclonal Antibody, AlexaFluor 647 conjugate (Product # MA3-39100-A647) at a dilution of 5 µg/mL in blocking buffer for 1 hour at room temperature protected from light. Nuclei (blue) were stained with Hoechst Dye (Product # 62249) at a dilution of 1:10,000 in blocking buffer. Images were taken on a Thermo Scientific ToxInsight Instrument at 20X magnification.

- Figure 4 Expression level of analyzed proteins of tight junctions in SKOV-3 and SKOV-3.ip cells cultured in monolayer and 3D in vitro models. ( A , C ). The distributions of SKOV-3 cells (left plot) and SKOV-3.ip cells (right plot) according to fluorescence intensity detected after staining with occludin-specific and ZO-1-specific antibodies (red--monolayer culture, blue--spheroids, green--collagen hydrogel); ( B , D ) Expression levels of occludin and ZO-1 in monolayer and 3D models denoted as relative fluorescence values calculated as a ratio of mean fluorescence intensity of cells stained with specific antibodies to mean fluorescence intensity of cells stained with antibodies of isotypic control. mnlr , monolayer; sph , spheroids, clgn ; collagen hydrogel. ""*"" indicates significant difference in RF level from monolayer culture (ANOVA, Holm-Sidak's multiple comparisons test, p < 0.05).

- Figure 4 Other markers of airway epithelial differentiation are shown in selected images from non-CF and CF organoids. ( A ) Top panel is a confocal maximal projection of beta-tubulin IV in F508del/G551D organoid with a 3-D reconstruction in the bottom panel that was sliced into to show cilia protruding. Scale = 50 um. ( B ) Cross-section stained with acetylated tubulin in a non-CF organoid. Scale = 50 um. ( C ) Light microscopy image of still cilia (arrows) in a non-CF organoid. Scale = 100 um. ( D ) uOCT image of still cilia (arrows) in a non-CF organoid. ( C , D ) Stills correspond with Videos S3 and S4 . ( E ) Organoids also expressed CFTR, as seen in this representative example from F508del/R117H-5T organoids; co-localizion with ZO-1, seen in a non-CF organoid, is shown in Figure S2 . Scale = 50 um. ( F ) FOXI1-positive cell (arrow) in an F508del/F508del organoid. Scale = 50 um. Maximal projection of tight-junction (ZO-1) staining ( G ) and a three-dimensional hemisphere demonstrating apical (luminal) localization in F508del/F508del organoids ( H ). Scale = 50 um.
