- KD of Cathepsin S was achieved by transfecting A549 with Cathepsin S specific siRNAs (Silencer® select Product # s3762, s3763). Western blot analysis (Fig. a) was performed using whole cell extracts from the Cathepsin S KD cells (Lane 3), non-specific scrambled siRNA transfected cells (Lane 2) and untransfected cells (Lane 1). The blot was probed with Cathepsin S Polyclonal Antibody (Product # PA5-47088, 1µg/mL) and Rabbit Anti-Goat IgG (H+L) Superclonal Secondary Antibody, HRP conjugate (Product # A27014,0.25µg/mL, 1:4000 dilution). Densitometric analysis of this western blot is shown in histogram where Fig. b corresponds to KD of 37 kDa band and Fig. c corresponds to that of 24kDa). Decrease in signal upon siRNA mediated knock down confirms that antibody is specific to Cathepsin S..

- Western blot was performed using Anti-Cathepsin S Polyclonal Antibody (Product # PA5-47088) and 37 kDa, 24 kDa bands corresponding to Cathepsin S was observed upon treatments in A549 cells. Whole cell extracts (30 µg lysate) of A549 (Lane 1), A549 treated with IFN gamma (50 ng/mL for 12hr) (Lane 2) and A549 treated with IFN gamma (50 ng/mL for 24hr) (Lane 3) were electrophoresed using NuPAGE® 4-12 % Bis-Tris gel (Product # NP0321BOX). Resolved proteins were then transferred onto a nitrocellulose membrane (Product # LC2001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1µg/mL) and detected by chemiluminescence Rabbit Anti-Goat IgG (H+L) Superclonal Secondary Antibody, HRP conjugate (Product # A27014, 1:4000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005).

- Immunohistochemical analysis of Cathepsin S in immersion fixed paraffin-embedded sections of human lymph node. Samples were incubated in Cathepsin S polyclonal antibody (Product # PA5-47088) using a dilution of 15 µg/mL overnight at 4 °C. Tissue was stained using the Anti-Goat HRP-DAB Cell & Tissue Staining Kit (brown) and counterstained with hematoxylin (blue).
