Western blot analysis of IGFBP3 using 30 µg of HeLa lysate. Samples were loaded onto a 12% SDS-PAGE gel and probed with an IGFBP3 polyclonal antibody (Product # PA5-29711) at a dilution of 1:1000.

Experimental details

Western blot was performed using Anti-IGFBP3 Polyclonal Antibody (Product # PA5-29711) and a 36 kDa band corresponding to IGFBP3 was observed across cell lines tested along with a uncharacterized band (*) at 70 kDa. Whole cell extracts (30 µg lysate) of T-47D (Lane 1), A549 (Lane 2), U-87 MG (Lane 3), MCF 10A (Lane 4), MCF7 (Lane 5), BeWo (Lane 6), A-431 (Lane 7) and PANC-1 (Lane 8) were electrophoresed using NuPAGE™ 4-12% Bis-Tris Protein Gel (Product # NP0321BOX). Resolved proteins were then transferred onto a Nitrocellulose membrane (Product # LC2001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (0.3 ug/ml) and detected by chemiluminescence with Goat anti-Rabbit IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036, 1:4000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005).
