- Western blot was performed using Anti-PIAS2 Monoclonal Antibody (OTI5B4) (Product # MA5-26974) and a 63-68kDa band corresponding to E3 SUMO-protein ligase PIAS2 was observed across cell lines and tissue extract tested except Mouse Prostate, Mouse Spleen and Mouse Brain which is reported to be low. Fresh nuclear enriched extracts (30 µg lysate) of HeLa (Lane 1), DU 145 (Lane 2), LNCaP (Lane 3), Jurkat (Lane 4), K-562 (Lane 5), PC-3 (Lane 6), and tissue extracts (30 µg lysate) of Mouse Prostate (Lane 7), Mouse Spleen (Lane 8), Mouse Brain (Lane 9), and Mouse Testis (Lane 10) were electrophoresed using NuPAGE™ 4-12% Bis-Tris Protein Gel (Product # NP0322BOX). Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1:1000 dilution) and detected by chemiluminescence with Goat anti-Mouse IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A28177,1:4000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using SuperSignal™ West Dura Extended Duration Substrate (Product # 34076).
